What is the Nano-Glo® Luciferase Assay System?

The Nano-Glo® Luciferase Assay System is a dedicated detection reagent for NanoLuc® Luciferase, a 19.1kDa monomeric enzyme engineered from the deep-sea shrimp Oplophorus gracilirostris. The system uses the Nano-Glo Luciferase Assay® Substrate to generate bright, ATP-independent, glow-type luminescence with low background, enabling sensitive detection in cell-based assays. It is widely used in molecular biology, cell signaling, gene expression, drug screening, and protein interaction studies.

How Does the Nano-Glo® Luciferase Assay Work?

The Nano-Glo® Luciferase Assay uses a simple add-and-read protocol. First, combine the Nano-Glo® Luciferase Assay Substrate with Nano-Glo® Luciferase Assay Buffer to create the working reagent. The reagent lyses cells and simultaneously provides the NanoLuc® Luciferase Assay Substrate so no separate lysis step is needed.

When you add the prepared reagent to samples expressing NanoLuc® Luciferase, the luminescent reaction produces a stable light signal. The emitted light has a half-life of approximately 120 minutes in standard tissue culture media, providing a wide measurement window.

You can measure the signal using any standard plate reader or luminometer. Luminescence output is linearly proportional to NanoLuc® Luciferase concentration across a million-fold dynamic range, allowing you to detect both subtle changes and strong expressors.

Simple add-mix-read protocol:

  1. Add the prepared Nano-Glo® Luciferase Assay Reagent directly to cells or conditioned media.
  2. Mix briefly.
  3. Measure luminescence on any standard plate reader or luminometer.

Can I use the Nano-Glo® Luciferase Assay with secreted NanoLuc® Luciferase?

Yes. The Nano-Glo® reagent can be added directly to conditioned culture media to detect secreted NanoLuc® Luciferase without cell lysis. This makes it suitable for monitoring secretion-based reporter assays or for serial sampling of the same culture over time.

 

Do I need an injector-equipped luminometer to run this assay?

No. The Nano-Glo® Luciferase Assay produces a stable glow-type signal with approximately 120-minute half-life, so there is no need for injectors or rapid-read instrumentation. Any standard microplate reader or luminometer capable of detecting luminescence will work, including the GloMax® Discover System.

 

How sensitive is the Nano-Glo® Luciferase Assay compared to other luciferase assays?

NanoLuc® Luciferase produces approximately 100-fold greater luminescence intensity than firefly or Renilla luciferase. Combined with the low autoluminescence of the Nano-Glo® Luciferase Assay Substrate, this enables detection of reporter expression at levels where traditional luciferase systems may produce insufficient signal-to-background ratios.

 

Can I use the Nano-Glo® Luciferase Assay for BRET experiments?

The Nano-Glo® Luciferase Assay System is designed for standard endpoint lytic assays. For BRET and NanoBRET® applications, we offer dedicated NanoBRET® Nano-Glo® Detection Systems optimized for energy transfer measurements with live cells.

 

Can I use the Nano-Glo® Luciferase Assay for live cell experiments?

The Nano-Glo® Luciferase Assay System is designed for standard endpoint lytic assays. For live-cell NanoLuc® detection without lysis, see the Nano-Glo® Live Cell Assay.

 

Is the Nano-Glo® Luciferase Assay compatible with high-throughput screening?

Yes. The single-addition, no-injector format and extended signal stability make the Nano-Glo® Assay well suited for batch-mode and continuous-process HTS workflows. The assay scales from individual tubes to 384-well and 1536-well plate formats.
